Video: Mini-lab tour for undergraduate organic labs

Our laboratory maintains five nuclear magnetic resonance (NMR) spectrometers.

Bruker Avance III HD 500 (HD-500)

Bruker Avance III HD 500 MHz (HD-500)

This Bruker Avance III HD 500 MHz is a 2-channel spectrometer with a shielded Ascend magnet and a 60-place SampleXpress autosampler. There is one probe available, a 5 mm Prodigy TCI-H&F cryoprobe (single axis Z-gradient). This instrument is typically run under full automation with IconNMR. The magnet sits on a Technical Manufacturing Corporation vibration table to protect it from light rail vibrations. The magnet and autosampler were installed in 2011, and the console and Prodigy probe were installed in July 2013. It is located in 194 Kolthoff Hall.

5 mm Prodigy TCI-H&F cryoprobe specifications (1H or 19F on the inner coil, 13C on an outer coil):

  • 1H S/N: 2500:1
  • 13C S/N: 350:1
  • 19F S/N: 2000:1
  • Temperature range: this spectrometer is not typically used for variable temperature work and it is normally set to 298 K.  However, the probe is capable of 0 °C to 135 °C.

Bruker Avance III 500 (AV-500)

Bruker Avance III 500 MHz (AV-500)

This Bruker Avance III 500 MHz is a three-channel spectrometer with a shielded Ascend magnet and a 24-place SampleCase autosampler. Two probes are available. The typically installed probe is a 5 mm BBFO SmartProbe (single axis Z-gradient); it has two channels: broadband (BB) and 1H. The second available probe is a 5 mm TBO triple resonance broadband probe (single axis Z-gradient); it has three channels: BB, 1H, and 19F.  The TBO probe allows acquisition of 13C spectra with simultaneous 1H and 19F decoupling: 13C{1H+19F}. Please contact NMR personnel if you desire a probe change. A BCU-1 chiller is available for decreasing the probe temperature to ~0 °C.  Temperatures below 0 °C are accessible through the use of a liquid nitrogen evaporator accessory.  This instrument is run in manual mode, but automation is available with the autosampler through IconNMR. The console and probes were installed in December 2011, and magnet and samplechanger were installed in 2013. It is located in 196 Kolthoff Hall.

5 mm BBFO SmartProbe specifications (BB on inner coil, 1H on outer coil):

  • 1H S/N: 730:1
  • 13C S/N: 250:1
  • 19F S/N: 550:1
  • 31P S/N: 180:1
